V dnešní době se používají výhradně sférické částice, které vykazují oproti starším částicím s nepravidelným tvarem vyšší účinnost. Velikost původně vyráběných částic byla několik desítek mikrometrů. Při plnění kolon vznikal velký prostor mezi částicemi, a tím docházelo ke snížení účinnosti separace vlivem zvýšené turbulentní difúze a zvýšeného odporu proti převodu hmoty1. Vývoj nových technologií umožnil přípravu částic s průměrem pouhých několika mikrometrů. Tyto částice mají vyšší účinnost a kapacitu, ale také vyšší tlak na koloně2. Nejčastěji používaný průměr částic je dnes 3 – 10 µm. Částice mohou být neporézní, mohou mít porézní povrchovou vrstvu, nebo být porézní.
Existence neporézních částic je důsledkem snahy o získání vysoké účinnosti separace při nižším tlakovém spádu na koloně. Absence pórů snižuje celkový odpor proti převodu hmoty eliminací příspěvku k odporu převodu hmoty v mobilní fázi stagnující v pórech1. Výsledkem je zvýšení účinnosti separace. Nevýhodou kolon plněných neporézními částicemi je výrazné snížení kapacity, které je důsledkem menšího povrchu částic3.
Částice s porézní povrchovou vrstvou se skládají ze dvou vrstev. Vnitřní vrstvu tvoří kompaktní kulovité jádro z anorganického materiálu, často velmi čistého pevného silikagelu. Neporézní jádro je překryté slabší vrstvou porézního materiálu. Čím tenčí je porézní vrstva, tím snazší je převod hmoty a roste účinnost separace. To je výhodou zejména pro makromolekuly, které difundují do a z pórů pomaleji než malé molekuly. Zároveň se ovšem s klesající tloušťkou porézní vrstvy snižuje specifický povrch stacionární fáze, a klesá sorpční kapacita kolony4,5.
Kolony plněné porézními částicemi jsou nejčastěji používané. Částice jsou z anorganického materiálu, jejich jádro je nejčastěji z oxidů křemíku, hliníku nebo zirkonu. Mají vysokou účinnost i kapacitu, ale jejich nevýhodou je značný příspěvek odporu proti převodu hmoty ve stagnující mobilní fázi k rozmývání elučních zón. Způsobem, jak lze tento vliv potlačit, je použití perfuzních částic s průtočnými póry, které vykazují nižší odpor proti převodu hmoty a umožňují použití vyšší průtokové rychlosti bez ztráty účinnosti separace6,7.
